AMR103 (TARGETED TRANSPORT OF LEVODOPA)
INTRODUCTION
Levodopa is the "gold standard" for the alleviation of Parkinson's disease symptoms, accounting for 70% of the prescription market. Currently marketed formulations of levodopa are poorly and erratically absorbed and extensively broken down in the periphery. Only 1% of the oral dose gets to the brain. Moreover, long term treatment with levodopa is associated with response fluctuations, dyskinesias and psychiatric disturbances.
AMR103 is a single chemical entity resulting from Amarin's Targeted Lipid Transport technology that chemically conjugates levodopa and a specific bioactive lipid. Amarin's targeted lipid transport program utilizes combinatorial lipid technology which chemically conjugates bio-active lipids with either other lipids or existing drugs to improve bioavailability, blood brain barrier penetration and potentially increase efficacy whilst reducing side effects. Each conjugate will be a new chemical entity with the potential for new intellectual property.
DEVELOPMENT TO DATE
AMR103 has been shown in preclinical studies to significantly increase brain levels of dopamine. This could potentially lead to potentially fewer doses per day reducing the peaks and troughs believed to contribute to a cycle of 'on-off' and/or dyskinesia. Further preclinical studies will be conducted before filing an IND and commencing a Phase I study.
